ABSTRACT

In the present study the effect of BMP-6 was investigated on chondrogenesis of adipose-derived stem cells. Mesenchymal stem cells derived from subcutaneous adipose tissue were cultured on alginate scaffold to induce chondrogenesis in experimental group, with chondrogenic medium having BMP-6 growth factor for 3weeks. In control group medium without BMP-6 was applied. The harvested constructs were examined with immunohistochemical and RT-PCR methods for assessment of cartilage-specific characteristics. The results of immunohistochemical method revealed the presence of typical cartilage extracellular matrix components such as type II collagen and aggrecan in constructs induced by BMP-6 growth factor on alginate scaffold. In addition evaluation of the results of RT-PCR analysis confirmed the expression of cartilage- specific genes, such as type II collagen and aggrecan, in the differentiated cells under the influence of growth factor BMP-6. It can be concluded that BMP-6 promotes chondrogenesis of ADSC in 3-D and adipose-derived stem cells could be used for cartilage tissue engineering

ABSTRACT

Cystic hyfroma is a disorder in lymphatic vessel formation that involves the adjacent organs and can affect them due to its fast growing nature. The main treatment for cystic hygroma is surgical intervention that an have many complications after surgery. Treatment other than surgical interventions is the use of sclerosing agents. The goal of this study is to analyze the effect of Beliomycin in treating cystic hygroma. This quasi experimental study was carried out clinically at st-al Zahra and Kashaine hospitals of Isfahan from 1372 to 1383. A checklist including age, sex, cyst location, cystic hygroma volume before and after Beliomycin injection and/or surgery, was completed for each patient. Data was analyzed by T and paired T-Tests. p< 0.05 was considered meaningful. The study was carried out on 72 patients. 24 patients were treated with Beliomycin and 48 patient by surgery. Patients age was below 1 months up to 18 years old. Cystic hygroma was mainly located at neck region [46.8%]. The average size of cystic hygroma was 103.9 +/- 29.66cm[3] before Beliomycin injection and 34.91 +/- 16.19cm[3] after that [p=0.004]. and 43.511 +/- 7.81cm[3] before surgery and 1.39 +/- 1.39cm[3] after that [p=0.006]. The rate of recurrence of lymphingoma in the surgery treated group was 25.1% and in the group treated with Beliomycin was 29.2% with no significant diffference. Regarding the considerable decrease in cyst volume after Beliomycin injection and it's low complication compared to surgery, this method can be used as a primary treatment or supplementary to surgery

ABSTRACT

To describe the outcome of the sapheno-femoral fistula as an alternative blood access site formaintenance hemodialysis in a prospective cohort of patients with end - stage renal failure.Twenty-two patients with vascular access failure in the arms were admitted for establishing saphenofemoralfistula as a puncture site for hemodialysis. The major saphenous vein was exposed at the junction sitewith femoral vein and its tributary veins were ligated. The saphenous vein was isolated and mobilizedthroughout the thigh and end to side anastomosis accomplished with the superficial femoral artery by a running6-0 prolene suture after conducting the vein through a subcutaneous tunnel.Failure rate of saphenofemoral fistula was 22. 7% at two years follow up. Mean +/- SE survival of fistulawas 16.4 +/- 2.75 months. Significant survival difference wasn't seen between two sexes. Rate of woundinfection and chronic pain of surgery site was similar [9.1%].Two-year survival rate of 77% and morbidity less than 10% leads to suggestion of saphenofemoralfistula as an alternative for upper extremity fistulas in end- stage renal failure patients
